Chang et al. (1986) isolated and sequenced a cluster of 4 tRNA genes: proline-leucine-spacer-proline-threonine. The 2 tandem pairs of genes are separated by 3 kilobases. A proline tRNA(AGG) gene is separated from a leucine tRNA(AAG) gene by a 724-bp segment, and a second proline tRNA(AGG) gene is 316 bp from a threonine tRNA(UGU) gene; the leucine tRNA gene has a polarity opposite to that of the other 3 genes. Olson et al. (1987) used portions of this segment as probes for chromosome localization of the cluster by Southern hybridization in human-rodent somatic cell hybrids. These studies demonstrated localization of the cluster of genes on chromosome 14. RFLPs detected in the cluster will allow determination of genetic linkage to other polymorphic loci on that chromosome. Mitchell et al. (1989, 1991) mapped the LPT tRNA gene cluster to chromosome 14 by somatic cell hybrid analysis, in situ hybridization, and linkage analysis. In situ hybridization placed the cluster at 14q11-q12. RFLPs detected by the tRNA clones and by TCRA genes (see 186880) were used to analyze CEPH pedigrees for linkage; a maximum lod score of 6.02 at 6 cM was observed for these 2 loci.
