▼ Description
Members of the NYAP family, including NYAP1, are predicted to have a role in neuronal morphogenesis (Yokoyama et al., 2011).

▼ Cloning and Expression
Yokoyama et al. (2011) cloned mouse Nyap1. The deduced 832-amino acid protein contains a central NYAP homology motif (NHM) that is shared with other NYAP proteins. The NHM is proline-rich and contains 2 putative phosphotyrosines. Northern blot analysis detected Nyap1 expression in mouse brainstem, cerebrum, and cerebellum, with much lower expression in small intestine and colon. No expression was detected in other peripheral tissues examined. Northern blot analysis detected Nyap1 in rat cortical neurons, but not in astrocytes or a rat oligodendrocyte cell line. In situ hybridization detected Nyap1 in the cortical plate on embryonic day 14. In postnatal mouse brain, Nyap1 was expressed in neocortex and striatum, but not in olfactory bulb. Phosphorylated Nyap1 had an apparent molecular mass of 100 kD.

▼ Gene Function
Using cultured mouse cortical neurons, Yokoyama et al. (2011) found that Nyap1, Nyap2 (615478), and Nyap3 (MYO16; 615479) were phosphorylated by Fyn (137025) following stimulation with the neural adhesion molecule contactin-5 (CNTN5; 607219). No other growth factor or signaling molecule examined led to Nyap phosphorylation. All 3 phosphorylated Nyap proteins interacted with the p85 regulatory subunit of PI3 kinase (see 171833) and activated PI3 kinase for generation of phosphatidylinositol 3-phosphate (PIP3). The Nyap proteins also interacted with the Wave1 (WASF1; 605035) complex, which mediates remodeling of the actin cytoskeleton. The simultaneous interaction of Nyap proteins with p85 and the Wave1 complex activated the Wave complex via PIP3 and activated Rac1 (602048), resulting in actin remodeling.

Hartz (2013) mapped the NYAP1 gene to chromosome 7q22.1 based on an alignment of the NYAP1 sequence (GenBank AK094857) with the genomic sequence (GRCh37).

▼ Animal Model
Yokoyama et al. (2011) found that triple knockout of Nyap1, Nyap2, and Nyap3 in mouse resulted in reduced brain size and stunted neurite elongation in culture.
