Methyltransferases catalyze the transfer of a methyl group from a donor, generally S-adenosyl-L-methionine (SAM), to acceptor molecules, including proteins, DNA, RNA, lipids, and small metabolites. METTL21A belongs to a family of protein lysine methyltransferases that methylate nonhistone proteins (Cloutier et al., 2013).
▼ Cloning and Expression
By searching databases for sequences similar to METTL22 (615261), Cloutier et al. (2013) identified METTL21A. The deduced METTL21A protein is made up predominantly of a Rossmann fold of alternating beta strands and alpha helices, with a conserved SAM-binding site near the N terminus. Epitope-tagged METTL21A was predominantly expressed in a diffuse cytoplasmic distribution in transfected HeLa cells.
▼ Gene Function
By affinity purification, SDS-PAGE, and mass spectrometry, Cloutier et al. (2013) found that METTL21A expressed in HEK293 cells interacted with HSP110 (see 610703) and HSP70 (see 140550). In vitro methylation assays showed that recombinant METTL21A methylated the HSP70 proteins HSPA1 (HSPA1A; 140550), HSPA5 (138120), and HSPA8 (600816). Mutation of the target lysines in the C-terminal tails of HSPA1, HSPA5, and HSPA8 abrogated their methylation.
Using a synthetic SAM analog for in vitro assays, Shimazu et al. (2014) showed that human METTL21A had methyltransferase activity for histone H3 (see 602810) and the HSP70 proteins GRP75 (606704), GRP78 (606921), and HSP7C. METL21A also had automethyltransferase activity.
Hartz (2013) mapped the METTL21A gene to chromosome 2q33.3 based on an alignment of the METTL21A sequence (GenBank AF455817) with the genomic sequence (GRCh37).
