▼ Description
IL1F10, or IL38, binds to IL36R (IL1RL2; 604512) and inhibits production of various other cytokines (van de Veerdonk et al., 2012).

▼ Cloning and Expression
By searching databases for novel IL1 (see IL1A; 147760) family members, Lin et al. (2001) identified a cDNA for IL1F10, which they termed IL1HY2, from a human fetal skin library. Using RACE, they obtained a full-length cDNA encoding a predicted 152-amino acid protein with a calculated molecular mass of 17 kD. IL1HY2 shares significant amino acid homology with IL1RA (IL1RN; 147679) and IL36RN (605507) and lower homology with IL1A and IL1B (147720). PCR analysis of various human cDNA libraries detected IL1HY2 expression in fetal skin and, at a lower level, in spleen. Recombinant IL1HY2 expressed in Chinese hamster ovary cells was detected as a secreted protein. Immunohistochemical analysis demonstrated expression of IL1HY2 in basal epithelia of human skin, in human tonsil germinal center, and in some proliferating tonsillar B cells, but not T cells.

▼ Gene Function
Lin et al. (2001) found that IL1HY2 bound soluble IL1R1 (147810) weakly compared with IL1RA and IL1B.

By stimulating human memory T lymphocytes with heat-killed Candida albicans, van de Veerdonk et al. (2012) found that the addition of low, but not high, concentrations of IL38 inhibited production of IL22 (605330) and IL17 (603149), similar to the reduction caused by IL36RN. Production of IL8 induced by IL36G (605542) was also reduced by both IL38 and IL36RN. IL38 and IL36RN caused monocyte-derived dendritic cells stimulated with lipopolysaccharide to double production of IL6 (147620). IL38, like IL36RN, bound to IL36R, but not to other members of the IL1 receptor family. Van de Veerdonk et al. (2012) concluded that both IL38 and IL36RN bind to IL36R and have similar effects on immune cells.

▼ Gene Structure
Lin et al. (2001) determined that the IL1HY2 gene has 4 coding exons, similar to other IL1 family genes.

▼ Mapping
Using radiation hybrid analysis, Lin et al. (2001) mapped the IL1HY2 gene to chromosome 2q14, close to other IL1 family members.

Nicklin et al. (2002) determined that the gene order within the IL1 gene cluster on chromosome 2, from centromere to telomere, is IL1A-IL1B-IL1F7 (IL37; 605510)-IL1F9 (IL36G)-IL1F6 (IL36A; 605509)-IL1F8 (IL36B; 605508)-IL1F5 (IL36RN)-IL1F10-IL1RN. Of these, only IL1A, IL1B, and IL36B are transcribed toward the centromere.
