▼ Cloning and Expression
By database analysis, followed by screening a human blood cell cDNA library, Zou et al. (2006) cloned RASL10B. The deduced 203-amino acid protein has a molecular mass of 23.2 kD and contains 5 guanine nucleotide binding sites and a C-terminal CAAX box. RT-PCR analysis of human tissues detected strong expression in brain, pancreas, and prostate, with moderate expression in lung and thymus, and low expression in liver and spleen. Immunofluorescence localized RASL10B to the cytoplasm in COS-7 cells. RASL10B mRNA was downregulated in human breast cancer cell lines compared to a normal human breast cell line.

Rybkin et al. (2007) used microarray analysis to compare mRNA derived from NIH3T3 fibroblasts and a mouse cardiac cell line. They selected ESTs expressed specifically in the cardiac cells and identified an EST encoding RASL10B, which they called RRP17. Northern blot analysis detected a single 3.6-kb transcript in human heart, brain, skeletal muscle, and pancreas, as well as mouse heart and brain. In situ hybridization of mouse embryos detected Rasl10b in developing heart, skeletal myotomes, and dorsal portion of the neural tube at embryonic day (E) 9.5. At E13.5 and E15.5, Rasl10b was detected in heart and central and peripheral nervous system.

By yeast 2-hybrid screen of brain and heart cDNA libraries, Rybkin et al. (2007) showed that RRP17 interacted with the C-terminal region of CAPS1 (CADPS; 604667), a regulator of large dense-core vesicle secretion. They confirmed this interaction by GST pull-down assay. Primary rat neonatal cardiomyocytes infected with recombinant adenovirus expressing RRP17 secreted 2.5- and 3.5-fold more atrial natriuretic peptide (ANP; see 108780) compared to uninfected or lacZ-expressing cardiomyocytes.

Zou et al. (2006) stated that the RASL10B gene maps to chromosome 17q12.

▼ Animal Model
Rybkin et al. (2007) generated Rrp17 -/- mice and found that they were viable, fertile, and obtained at the expected mendelian ratio. There was no obvious phenotype, but atrial cardiomyocytes from Rrp17 -/- mice showed large dense-core vesicles that were smaller in size compared to wildtype and decreased secretion of stored ANP. These mice also had significantly higher systolic and diastolic blood pressure.
