Micro RNAs (miRNAs), such as MIR574, are a class of 20- to 24-nucleotide regulatory RNAs that negatively regulate gene expression by binding target mRNAs. The MIR574 stem-loop sequence encodes 2 mature miRNAs, MIR574-5p and MIR574-3p, from its 5-prime and 3-prime ends, respectively (Tatarano et al., 2012; Wang et al., 2012).
▼ Gene Function
Using quantitative stem-loop RT-PCR, Tatarano et al. (2012) found that expression of MIR574-3p was downregulated in clinical bladder cancers compared with normal bladder tissue. MIR574-3p was also significantly downregulated in 2 bladder cancer cell lines. Gene expression analysis showed that the MESDC1 (615466) transcript, which contains a functional MIR574-3p-binding site, was significantly downregulated by overexpression of MIR574-3p. Knockdown of MESDC1 or overexpression of MIR574-3p reduced cell proliferation, migration, and invasion and induced apoptosis. Tatarano et al. (2012) concluded that MIR574-3p has a tumor suppressor function.
Wang et al. (2012) found that high serum MIR574-5p concentration was associated with survival from sepsis.
