▼ Cloning and Expression
Many human tumors express antigens that are recognized in vitro by cytolytic T lymphocytes (CTLs) derived from the tumor-bearing patient. The MAGE (melanoma-specific antigen) and GAGE (G antigen) gene family members encode such antigens. Chen et al. (1998) manipulated the androgen-sensitive, nontumorigenic, prostate-specific antigen (PSA)-secreting cell line LNCaP in vivo to produce 3 cell sublines that were progressively more tumorigenic, metastatic, and androgen-insensitive. To screen for genes associated with cancer progression, Chen et al. (1998) performed differential display PCR using these sublines and isolated 2 members of the GAGE family, GAGEB1 and GAGE7 (300601). GAGEB1 encodes a 143-amino acid polypeptide that contains 6 potential casein kinase phosphorylation sites, 1 potential protein kinase C phosphorylation site, and 3 potential N-myristoylation sites. Northern blot analysis revealed that GAGEB1 is expressed in the placenta and testis. Levels of GAGEB1 mRNA were correlated to the degree of tumorigenicity of prostate cancer cell lines, but were unaffected by androgen treatment.

By searching EST databases for sequences preferentially expressed in adult reproductive tissues, Brinkmann et al. (1998) identified PAGE1. Northern blot analysis detected a transcript of about 500 nucleotides expressed predominantly in placenta, fallopian tube, and prostate, and at lower levels in testis and uterus. It was also expressed in uterine cancers but not expressed in other normal or cancer tissues examined. RNA dot-blot analysis and RT-PCR confirmed expression predominantly in reproductive tissues.

By Southern blot analysis and genomic sequence analysis, Brinkmann et al. (1998) mapped the PAGE1 gene to chromosome Xp11.23.
