▼ Description
DNAJB5 belongs to the evolutionarily conserved DNAJ/HSP40 protein family. For background information on the DNAJ family, see 608375.
▼ Cloning and Expression
By searching for sequences similar to bacterial Dnaj, followed by PCR amplification, Chen et al. (1999) obtained a partial human DNAJB5 clone, which they called HSC40. They cloned full-length mouse Dnajb5, which encodes a deduced 462-amino acid protein. Northern blot analysis of human tissues detected ubiquitous expression of DNAJB5, with highest levels in heart, followed in order by skeletal muscle, spleen, liver, brain, and pancreas, and very weak expression in lung and kidney.
▼ Gene Function
Chen et al. (1999) found that the expression of DNAJB5 in normal human fibroblasts was relatively high under normal growth conditions and moderately induced by heat shock or sodium arsenite. This expression pattern is similar to that of HSC70 (HSPA8; 600816) but different from that of HSP40 (DNAJB1; 604572), which is low under normal growth conditions and significantly induced by heat shock or sodium arsenite.
▼ Gene Structure
Chen et al. (1999) determined that the mouse Dnajb5 gene contains 5 exons. The promoter region contains a heat-shock element and a TATA box.
▼ Mapping
The International Radiation Hybrid Mapping Consortium mapped the DNAJB5 gene to chromosome 9 (RH62785).
