GGT6 belongs to the gamma-glutamyltransferase (GGT; EC 220.127.116.11) gene family. GGT is a membrane-bound extracellular enzyme that cleaves gamma-glutamyl peptide bonds in glutathione and other peptides and transfers the gamma-glutamyl moiety to acceptors. GGT is also key to glutathione homeostasis because it provides substrates for glutathione synthesis (Heisterkamp et al., 2008).
▼ Cloning and Expression
By searching human genomic and cDNA databases for sequences related to GGT1 (612346), Heisterkamp et al. (2008) identified GGT6. The predicted GGT6 mRNA is 2.5 kb, and the deduced protein contains 461 amino acids. GGT6 shares limited amino acid homology with GGT1. EST database analysis suggested that GGT6 expression is restricted to esophagus, trachea, kidney, bladder, adrenal gland, and intestine, as well as adrenal, colorectal, and breast carcinomas.
▼ Gene Structure
Heisterkamp et al. (2008) determined that the GGT6 gene contains 3 exons and spans less than 5 kb.
By genomic sequence analysis, Heisterkamp et al. (2008) mapped the GGT6 gene to chromosome 17p13.2.
