▼ Cloning and Expression
Moreno et al. (2001) cloned human and mouse MOB1. The deduced 225-amino acid mouse and human proteins are identical, and the cDNAs share 95% nucleotide identity. Moreno et al. (2001) determined that MOB1 is the class II homolog of yeast Mob1. Immunofluorescence analysis of NIH 3T3 mouse fibroblasts detected Mob1 in a punctate distribution throughout the cytoplasm, with high concentration close to the nuclear membrane. Mob1 colocalized with striatin and S/G2 nuclear autoantigen (Sg2na). Western blot analysis detected MOB1 as a 24-kD doublet.
▼ Gene Function
Moreno et al. (2001) found that inhibition of protein phosphatase-2A (PP2A; see 176915) in NIH 3T3 fibroblasts induced phosphorylation of PP2A-associated mouse Mob1 on serine, as well as hyperphosphorylation of striatin, Sg2na, and 3 other proteins. Immunoprecipitation analysis of NIH 3T3 cells detected about a third of endogenous Mob1 in a complex with striatin and another third associated with Sg2na. Moreno et al. (2001) concluded that MOB1 may be a PP2A substrate that has a role in modulating membrane-cytoskeletal interactions.
Gross (2011) mapped the MOBKL3 gene to chromosome 2q33.1 based on an alignment of the MOBKL3 sequence (GenBank AB015441) with the genomic sequence (GRCh37).
